You can, just be sure to provide some land for them to crawl on, the correct frozen/live foods, and warm humid air above the water. The salinity can be anything from 1.006 and upwards, just not pushing marine conditions too much though.

I've had mudskippers before and they are actually pretty easy to maintain as long as you provide the necessary special arrangments involving land portion for them. Other than that, the will accept most foods, I would alternate between NLS pellets, frozen krill, and frozen bloodworms

I think it's worth mentioning that with a 29 gal you're much better off with a small species like the dwarf Indian mudskippers. Large 'skippers like Africans (the most common) are aggressive and would probably outgrow that tank. With dwarves, you could probably have more than one. I'd say 4 or 5 for that tank max, and make sure there are multiple places on land for them to claim territory.
Also, as with most gobies, it's best that you use a fine sand substrate, to avoid damaging their soft skin or fins. They'll probably want to burrow, so something they can dig in is best.

How deep will the water be? I would probably use a sponge filter because a filter or pump that uses an impeller that close to the substrate would likely suck up grains of sand, which would destroy the impeller.
Frogman- Link below to a build i just recently finished. With a 29 this style may not work but you may pull out an idea or so. Good luck and post pics with update when done!

Mud skippers are fun- I had 2 dwarf skippys for a long time in a 20L. At first I had it as a beach set up, and they didnt use the water much, so I switched to stacked rocks. I ended up with 2 males and this seemed to suit them best because they could inhabit thier own slate piece; driftwood is also nice. Mine loved bloodworms, but their favorite was flighless fruit flies, weevles and house flies (dewinged). I had a Green Spotted Puffer living the the water with a couple of zebra danios and later 2 skunk botias (snail infestation)
