Depends on the 2x3's. If they are your typical kiln dried spruce it will be fine provided you build it properly. Sometimes 2x3's are finger-jointed though, in which case they have very little strength comparatively.

Depending on the grade of the lumber you should be fine. If there are minimal knots and long cracks going along the grain you should be fine. 45 gallons of water is about 375 pounds. Then you add in the tank and any ornaments/gravel going in so maybe 450 pounds. It would be spread to the corners of the stand mostly depending on your design, so about 110ish pounds per corner.
