Unless you want to use them as canisters, that's the best design.

1/4" tube will jam, eventually, 3/8" is the minimum that I would use. On the aerobic side.
1/2" on the anaerobic 100 Gpd, maybe bigger. I don't really know for sure.

Pvc glue is not a necessity below 15 psi. Nice to have removable plumbing.
