id like to know roughly how man lines I can run and how to set it up to run multiple tanks on 2 floors.

I picked this up for 35$ Canadian.

Post the brand, model and wattage of the pump, I can't see it clearly in the video.

You can use either PVC pipe or vinyl hose to split the air pump output between floors. Ideally you will want the air pump on the uppermost floor above the aquarium height to avoid siphoning any water into the air pump.

How do I cap vinyl hose? And what do I use to tap into with air lines for sponge filters
 
Vinyl hose can be plugged using barbed fittings that are capped or blind. I've seen them available at Home Depot or similar stores.

You can buy individual air valves that are threaded on the ends and drill and tap PVC pipe to accept the valves. They probably won't work for vinyl hose though.

Most people create a manifold using PVC pipe to distribute the air in fish rooms but this can become more complicated when having tanks on 2 different floor levels in a home.

Do you have a way to run vinyl hose or PVC pipe between the different floor levels? Some people drill a hole through the floor or walls and snake the necessary hose/pipe up and down. It gets a bit easier if the tanks or rooms are located above each other instead of at the opposite ends of the house.
