Can I increase the drain by doing this?

On my overflows I have the adjustable pipe the slides up and down. On the top is a small hole for air to vent. Could I drill this air vent hole and make it bigger? Would doing that allow for more water to drain by giving it more air flow? I definitely have the room in the drain to the sump for more water.

I don't believe so, if you're talking about the pipe I think you're talking about, all it needs is a hole, regardless of size, to allow air to escape.

Size shouldn't make a difference.

Only if the air is restricted. If it isn't "hissing" at all it probably won't make much difference. By the way, it should be sucking air in, not blowing out.
