So I want to cycle my sump before I put the water in my 65 gal, because I still need to do my auto top off before I fill the top tank which makes my sump inaccessible (Because doors arent big enough on stand for me to remove sump). Would I just run my hobby pump into my filtration and just make it cycle like that, and is heat necessary while there is no fish yet? Does nitrifying bacteria need heat?

No, they don't need heat (after all, people keep goldfish in unheated tanks, and the bacteria are still there), but heat does help them multiply faster. Not much faster, it's still going to take over a month to cycle... but somewhat faster. But it's okay if it isn't heated.
