Dechlorinator/ammonia removers affecting ammonia test?

I was told by a LFS a long time ago that some dechlorinators/ammonia removers can affect the results of some ammonia tests. It was only supposed to be an issue if the test was done shortly after the use of the dechlor/NH3 remover. Is there any truth to this? In an established aquarium is it more common to get ammonia spikes than nitrite spikes? Got some strange readings on my most recent test.
 
If it matters, I am using the API ammonia test with the two liquid reagents (not dip strips) and Prime ammonia detoxifier and dechlorinator.
