At what size does plywood become cost effective versus buying a large used setup?I'm thinking of ether building something over the next year or just saving up and buying a used setup.Labor and equipment for the plywood tank wouldn't be a problem my uncle is a carpenter and builds furniture as a hobby sometimes so he would help me with some of it.

Depends on the specific materials in which you want to build the tank. But.... typically it is somewhere in the 300-400 gallon range.

Yea I've researched some and love the idea of building my own aquarium.I'm gonna keep reading up and figure out what I wanna do for sure.I'm thinking around a 500 gallon if I have the space for it.The main thing I think about is durability of a diy tank.

Cost wise I agree with Pharaoh, however satisfaction of a job well done, and having a tank that nobody else in the world has worth the extra cost in a smaller size. I built a 240(ish) for around $1000 including stand, lighting and custom filter.
