Just finished glueing up my new sump to my Aqueon 75 gallon aquarium.
How long should I wait until I run water through the system for leaks?

Whoa whoa whoa...are you saying you just glued the plumbing together? Or that you built the sump itself from glass panels and silicone? If you built the sump tank from scratch its one week at the least.

No leaks! Well, one leak under emergency drain.
Bulkhead just needed to be snuggled up. Also the union below that had a slight drip. Opened it up dried it off and reset it and snugged it up.
no leaks since.
a few things I need help with.

even with emergency drain above water, it trickles due to the pipe not being glued into the overflow bulkhead. Should I glue it?

second thing is I can’t get the water vortex to go away in the main siphon. So all I hear is air coming through the main drain. Any recommendations would be great.

below is what I have
800gph Fiji cube overflow box
Hygger DC pump 1060gpg turned down to 58% at moment
I do have a nice gate valve at main drain for fine tuning .

You can wrap the standpipe with a thread tape to keep it from sucking at the bulkhead. That might solve your noise problem too.
The Fiji has 2 1" drains. So it doesn't have an emergency. A siphon and secondary is the best you can do. You'll want to start fiddling with the gate valve about half way with that pump and drain size to get it to raise the water in the overflow just enough to trickle into the second higher pipe.

I put a coarse prefilter (ac110 sponge block) over the full siphon drains in my fiji overflow box to mitigate the slurping sounds. Worked like a charm and creates an easily accessible prefilter.
