Could i keep a Fresh water Puffer fish of some sort with a FRT?
These would be the only occupants of the tank.

that's something that frustrates me very much. I really want a mbu puffer with my FRT but there's just no way I would risk keeping them together.
when it comes to compatibility questions such as this, it's a combination of more or less common sense and trial and error.
sometimes you just have to try it out in order to know the result. sometimes it works sometimes it doesn't.
HOWEVER, when it comes to a puffer fish and a turtle, it's common sense that tells you NO.
that puffer will nip, no questions about it. one day it will happen and you'll be really sorry when you come home to find an injured and possibly bacteria/fungus infected or worse yet, a dead FRT in your tank.

Or the FRT will bite the turtle, get a mouthful of puffer poison, and croak...probably a bad combo...
Definitly wouldn't risk it. The chances of the FRT getting bitten and then a fungal infection occuring are too high.
