i just bought a 180 gallon aquarium which came with a wet dry filter. When i set up the filter the first time a couple of the connections were loose and they leaked. so i went out and got some teflon tape and tube tighteners for the tubes. i set it up a 2nd time and .... it leaked again though not nearly as bad. any suggestions on how to fix theis problem?

silicone/pvc glue for the threads.. hose clamps and silicone for the tubes (hoses?)

I agree with Chesterthehero, but depending on what type of connections you are speaking of, you might be able to just replace worn/bad gasket material. More specifics always help. with hoses, just get some hose clamps.

Got any pictures of the area that's leaking?

NEVER put pvc glue on threads of ANYTHING. The threads are your best friend when the time comes to take something apart and put it back together. Only use silicone or teflon tape on threads. (Pipe dope is one of those things that should be left to people with experience using it.)
