Will my bichir/bichirs be able to survive in plastic bag filled with oxygen for 4-5 hours in a air-conditioned bus?
 
Yes, but I would put them in a cooler so that the water doesn't get so cold. The bichirs will come to the surface to get breath when they need to. How many bichirs and what size will determine how many bags you will need.
 
I thought air-conditioned always cools stuff?
Btw,what's a styro cooler?

Styro is short for styrofoam. A lot of small white foam pellets heat formed into an insulated box. Not sure what they would call it where you are but any insulated container should work to keep the water temp as close to normal as possible during the move.

Is it a must?
 
Not a must but it is less stressful on the fish. If no insulated container is available you can put them in a cardboard box and surround them with newspaper for insulation. If you can't do that make sure to acclimate them very slowly and very well before puting them in the new tank.
