what is the size of the spot? is it layer over the skin or sunken into the skin? any visual description of the texture of the affected skin area? any abnormal behavior like rapid breathing and scratching over wall or objects? any close up photos?

From what I hear, it may be wasting disease, fish TB, physical damage, sliminess of skin or ulcer disease. I cannot determine as the details are rather vague. I do advise isolation to treatment tank and proceed with low dose of broad-spectrum if the fish appears to be in a very bad state. I would use amocla (amoxicillin with clavulanic acid), it would be a little tough to get that for fish use, you may try nifurpirinol. This is only as a last resort. You should wait till the exact disease can be identified before proceeding to treatment.

Usage of diatom filter for sterilization is inefficient. The best way is to decomission the tank and use bleach to disinfect. Of course, UV would seem to be an easier choice. I usually remove all living stuff from the tank, deploy as many ozone generators and crank them all up for 2 hours, making sure I clear the premises and ventilate all areas. Then proceeding to soak it in bleach for 24 hours, rinse thrice and dry it with compressed air.
