I just got a 150 gallon long and I need to know if the floor needs to be reinforced if the tank is right next to a main wall of my house?

Depeneds, is it running paralell to or perpendicular to the support boards?

If its only sitting on 2 support boards (Paralell) that is bad, you need to reinforce.

It its perpendicular, you will be fine.

I have done the reinforcements and can show you pics of how to do it.

Will the tank set perpendicular to the floor joists?

i did. go under house and along side the floor stringers, pound in some more. i belive there 2x6's

i don't need it, but i'd be curious to see how you did it.

is there like a mathematical way to find out how much a tank would weight full of water?

If you go 10 pounds per gallon for the complete set up, it is a good estimate.

Otherwise if you want to be more accurate, you will have to calculate everything out separate. Water weighs 8.33 pounds per gallon.
