Is there any physical way of determining male/female? Or do you have to vent them?

You'll have to vent them or wait for a spawn. Males tend to be larger and have a more blocked forehead but a dominant female could easily fool you, and vice versa.
Ok the one is significantly bigger than the other and keeps chasing it, just thought they may be trying to mate
