Is silicone strong enough to seal a overflow in a acrylic aquarium?

No I would never use silicone for acrylic. Instead get some Weld-On 16 Acrylic Cement. You can get it cheap on ebay or amazon and it forms a permanent seal.

Weldon #40 is 30 times stronger than #16...says so right on their website. Most all tanks are built with #40. #16 is good for small repairs but it is brittle, full of bubbles and can start breaking down after a few years. Silicone does not adhere to acrylic so thats a no go. For the job ur doing id order a pint of weld on #40 or a small kit of #42. #42 is the same as #40 but comes in an epoxy type applicator that mixes it for you. Either glue should cost around $16-40. I think #16 is junk, i used it for years and watched it fail, later re-repairing my old work with #40. People on here swear by #16 but its cheap, readiliy available and they just dont know any better, its not their fault lol....

If it needs to hold water, no. If it is to attach an internal overflow box it would probably be fine, but using acrylic cement would be much better. The only benefit I could see is if you want to remove the weir in the future you could pull the silicone loose.
