I built a 35g out of someones 5/8" glass breakfront that they threw out, and the result holds water but is as ugly as can be. (will post pics later - it's outside and it's night.) I learned a few lessons.

1. Do it all in one shot. -don't tape up the silicone and come back later.
2. Use really long strips of tape (6"+) - short ones won't hold it together.
3. Cut the smallest hole in the tube nozzle you can - the smaller the stream the better.

I now have 55 & a 20 to reseal, and a lot of random glass to play with. Maybe i'll make a 6' x 8" x 8" tank - it'd be cool.

I would like to know if there any secrets to getting the silicone or cutting right. Tips, pics and good tutorial videos would be very much appreciated. :)

Setting the tape correct is key. Don't settle for the tape is slightly off straight, or it ripped after I got within an inch of the panel. Get it perfect laying the bead is less important but you should pay careful attention to the spread, having a caulk spreading tool will make the job much less messy. And of course as you noted go for it in one go, which includes up to removing the tape. Finally I don't care what the tube says about setting time its ALWAYS longer, I leave it at least a week. Also keep ventilation in mind, nothing worse then making the whole house smell like vinegar.
