Would this stand support a 29gallon tank or would it need to br much stronger?

It looks like that it is made out of MDF (vendor says it shouldn't be used in an area with condensation). I wouldn't use it. Besides, when they asked the vendor if it was strong enough to hold a PC, the vendor said no. A 29 gallon tank is roughly 240 lbs when filled, much heavier than a PC.

Would a basic frame made of 2x2" and 1" ply work?

I would suggest "2×4" and plywood.
