When to add water conditioner?

I have one of these Python type water changing things u hook p to a kitchen sink. My question is wen do i add the conditioner, before i add water or after? My old way i would fill a 5 gallon bucket with clean water, add conditioner, then add the water to the aquarium. I just want to be safe.

What if im only changing like 30 gallons? Put in enough conditioner for 60 gal?
 
From what I've seen that other people have said and what the back of the bottle says if you add the conditioner directly to the tank you have to dose for the whole tank. Thats why I stopped using my python now I just use it take water out of my 150 and I use a 4 gallon bucket to refill it. I use prime so I add 8 drops from a eye dropper to the 4 gallon bucket then put the bucket in the tub and fill it up and the water stirs it up.
 
FWIW I am no expert, but I drain and fill my 125 and 75 with a python and add conditioner as I refill. However, never read or knew you were supposed to dose for the whole tank. I have always added conditioner for the amount of water removed when refilling, never had any ill effects... Lucky?? IDK
