Nitrite in tap water and its known to cause birth defects in humans if levels above 10ppm are consumed by pregnant women. Not 100% of the time, but it's happened enough for them to know it causes it

That's from an EPA report from 2007. Nitrites cause methemoglobin-related health concerns in humans. Some cited are; Blue-baby syndrome, SIDS, retarded fetal growth development, cardiac defects, and nervous system defects.
