Bacteria needs air to survive, the more air in the sump the better.

　
Bacteria needs Oxygen to survive... and we have "Dissolved Oxygen" in our aquariums water to begin with, or else our fish would suffocate.
　
Utilizing the "wet/dry" approach to biofiltratoin is an option, but it is not a requirement. If it were a requirement canister filters would be obsolete...

how do you make refugium?

　
The basic concept of a refugium is to have plants in your sump… low light plants tend to reply more on nutrients from the water… fast growing plants tend to use more nutrients…
　
I suggest spending some time with Google and getting familiar with the concept of a freshwater refugium before making a decision wither way.

here is my 4ft sump with refugium. i added 500 bio balls to it. but it is still sitting on the floor with no water in it

I have a 50 gal sump with two huge bio-wheels, mesh filter media, etc. Super expensive but highly effective. Worth every penny.
