Yes, that is exactly how I was picturing it. Except I thought about cutting a 1/2 inch deep groove in the plywood bottom filled with silicon. Then I would Supported all around like you said with 2x4's for added security and bottom bowing prevention. I think I will still have a bar across the top because I am going to build a canopy for it. so you wouldn't see it anyway.
Sounds like you have a good plan. Acrylic is very flexible and the top is going to need as much, maybe more support, as the bottom for "bowing prevention" Check out any acrylic tank and the top piece is very thick and the openings in the top are small. My 360 only has 1/2 inch acrylic on the bottom but the top is 1" thick! Your going to need significant support for your top or the tank is going to rip apart.
