Presuming oxygen exchange is good, about 1/2 of the bacteria will die in 24 hours. Open the bag up, exchange the air in it, and shake the water around once in a while.
In a 3 hour drive you have nothing to worry about. I hope the pretty lady enjoys her new tank.
