Crack in floor - could it be from aquarium?

not from the tank from something else like settlement i know im in the industry and i have a 240 gallon on the secound floor but before filling i would deff try to see whats up underneath the floor before filling but more than likely it is do to a nail left behind under the floor and it is finally catching your eye but has been cracking for a while now a lil bit each day
 
I would agree that the increase in the width of the crack was due to the water, if it touched it, or the boards just settling. I would think you just haven't notice until now.

As far as the floor joists go, you should be OK, but I would keep an eye on things to make sure. 225 is a big tank to not have on a concrete slab. After you fill it, I would make a routine out of verifying that it is not too much weight by marking your beams and watching for movement.
 
Depending on the height of the basement, I would put some 4x4's for support underneath the tank just for piece of mind... As for the hardwood seperation, I also work in the flooring industry, and my thoughts are that it was already there and your concerns about the weight of the tank brought it to your attention. If the tank empty weighs 600 lbs three guys standing there would have caused the boards to separate which I dont believe is the case... The 4x4 legs under the tank would be a very cheap solution that I think would be worth their weight in gold if they prevent a huge hole in your 1st floor. GL and happy fish keeping.
