Very nice build man! Well thought out. A properly built floor with the tank up against the wall like it is will be fine. Middle of room is a different story. Also as long as the weight is spread out evenly among the joists you'll be fine. A floor is just a giant replica of a Diy tank stand lol.
 
Also that silicone is fine. Joey Diy uses it for all his builds. Including his 6x6 plywood bottom. I personally like the extra tensile strength of GE SCS structural silicone. Something you might like also.
