The green verticals do not need to be flush at the top or the bottom. They are only stiffeners to help in the framing process. They are not meant to touch the tank or the floor, according to the original designer.
The blue horizontal board in the top center can be flush with the top frame. Depending on the stand dimensions, you may need more than one.
I used that design for my six foot long 125G glass tank build and I love it. I also put a floor in my stand so added four additional 2x4's on their flat face (not on edge) to allow for inserting a 1/4" lauan panel. I caulked all the floor seams then painted with a few coats of a quality exterior white paint in case of minor water leaks when servicing my canister filters.
