Do I have to add water conditioner if I let the water sit for 2-4 days?

Either way, it is optional. You don't need to use it if you know your water is free from chemicals, etc that would harm your fish.

I meant that adding water conditioner and using a python is a lot easier and better than having bottles of water sitting around for the next water change.
