If you have a leaky glass tank, you need to peel out the silicone from all the corners (fresh silicone will not stick to old silicone). Clean the tank and corners with a decent solvent like alcohol and a razor blade to get all the bits of old silicone out. You shouldn't need to seperate the corners, just peel out the fillet bead of sealant. Then, get some GE silicone I - window and door, without mildew additive, or else get some silicone specifically labelled for aquarium use (anything else has toxins in it). Run fresh beads in all the corners, and let it cure for a few days (The tube will say 24 hours - wait longer)
It's not a huge job, but you can't just gunk on silicone over the old bead and expect it to hold up.

Cut the old panel out, yes. Clean it THOROUGHLY - any old silicone will weaken your joint. don't "gunk some silicone on the rims"... carefully put a uniform bead around the rim, avoiding air bubbles. Set the new panel in place and using tape or braces, or gentle clamps, hold it there for a few hours, until it's set enough to handle. Clean off any smeared or spilled silicone from the inside corners and run a bead around the inside, and use your finger to make it a nice even radius all the way around.
