Do a search on fx5 media and you will find all kinds of opinions on what to put in the different baskets.
 
indeed! I vote fine filter then bio all the way but its entirely up to your needs. Just remember the pre filter(the sponge on the inside of the FX5 is very very effective it has a ton of it so a lot of people choose to use mostly bio inside.With the exception of a few fine filter pads here and there. But do some looking around and find whats best for you.
 
Id say bio but that is just me.

There is a large and effective foam pre filter. If your worried you could put a pad in the bottom tray they tend to clog very quickly but in my experience that just means the filter is doing its job. It ultimately comes down to how often you can maintain it (Open it up and change out fine filter pads) If you go too long with the fine pads flow starts to really decrease.
