Have been planning a tank build for a while and just came across some glass

It looks good. The only thing I would change is to make the front a solid stacked 2x4 frame.

Horizontal below the glass, vertical besides it? basically fill in the voids? same pattern as it is now?
 
I would put all your external supports horizontally not vertically, think about the plywood, its most likely to bow on the longest plane. Alot of ppl di the vertical supports and they seem to work but I have to wonder how much they help.
We aren't building houses that have pressure pushing down on the walls, we have pressure pushing out on our walls therefore IMHO I would put them horizontal

Yeah, you want the area holding your glass in place as solid as possible.

The only thing about framing the studs horizontal is that the lower studs would have all the pressure of the water on them, spanning the entire length of the tank. With a vertical design only the lower parts of the studs have the deeper pressure on them. Maybe a better way to fix it would be to add some form of bracing to the bottom part. On my build I used the vertical stud design and then add a 2x6 to the lower part.
