I wouldn't trust that. I would over lap the hole by an inch or so. Are you going bare bottom? If not, your patch will be completely covered with sand if you use that. Also 1/4 inch acrylic will work fine to cover your holes. Hell the holes are already there you might want to hook up a closed loop system just to move the water around a bit more, that is a huge tank. A closed loop system with no filter or anything will be virtually maintenance free.

what everyone is saying about welding is the best way
altho you do not want acrylic squares in the open to cut your rays, id recoment using a big piece or putting silicon arounfd edge and or using substrate over it
 
I'm in awe!!! Those are gonna be some happy rays? What type of filtration will you be going with?
