Cool barn! Being from MN and spending a lot of time in WI, I love old barns.

You're going to need to insulate the heck out of that building. You're going to need a good insulated service door for starters. Then you're going to want to insulate the walls, and ceiling and probably frame up a sub-floor with insulation under it. The cheapest way would be gluing foam to the walls and window. If its feasible you could have someone come in a spray the whole thing with foam and be done with it.

Your also going to want to run or at least plan your electrical wires and water plumbing now before cover every thing up.

Are you going to put in a hot water heater? If not how do you plan to do water changes? The water will be COLD without a water heater.

As far as heat, I'd go for electric heat. That way you can seal the room up tight and not have to worry about ventilation and carbon monoxide. Plus electric heat is cheap to buy and with rising gas prices, probably cheaper to run.
