I have only had my precious MBU puff up once with air during a move. It was very slight as she started to make a weird noise when I netted her and transferred her to a bag with water for acclimation to her new enclosure.
I almost had a panic attack thinking she was going to fill up the net and get stuck. But as soon as she hit the water in the transfer bag she quickly deflated.
I would not push my luck with that.
 
When puffers inflate with air and not water for defense purposes, they have a high change of not being able to expel the air and thus dying while being stresses as well. General rule is NEVER have a puffer of any kind out of the water at any time.
