preparing for large bioload

Are you going to be running a canister or a sump? If your running a sump, I would divide it in have, add a circulation loop with a good size pump, and run a reverse flow sand bed. With 1.5 to 2 cubic feet of sand you can pack that tank in just a few days.

I use Bio Spira when I set up a tank that i won't be seeing everyday but the stability is 1/4 th the cost but you have to add it everyday for 1 week.
 
What type of fish are you adding?

I would just use old bio-media and cut my feeding schedule down to 1/4 for about 2-3 weeks and slowly re-build the bio-load.

Larger Cichlids can go a long time without food, as can most predatory fish. Feed them in small nutritional intervals, and you shouldn't spike your ammonia. If your worried about bacteria levels, I am a fan of 'Stability'; by Seachem.
 
It'll prolly be cichlids.

Actually, my situation has changed. So i'll probably fishless cycle the new tank on it's own and keep the old fish in their current tank. I'll jump start the cycle with old media and use the ammonia method.

For cichlids, go with my reverse flow sand bed and use aragonite sand. The aragonite will slowly disovle with the ph flucuation created by day/night temp differences. This will keep your ph and Alk (hardness) just where you need them to be.
