I would make sure your salinity is where you want it before adding live sand, wouldn't want the hyposalinity to kill whatever you've got living in your sand. You don't need to wait a day to check your salinity, just make sure it is well mixed. I usually put mine in a bucket with a pump going to mix, and when it's completely clear I'll test it.
