Yes it will help reguardless which way you use it. Right now, waste is accumulating in your gravel. It is causing an anaerobic bacteria colony to thrive. Unfortunately their byproduct can be toxic to fish if in high enough numbers (I killed the fish in my 18,000g swimming pool by turning on my dormant filter with about 150g of stale water (lots of dead algae, pond scum, etc., and lots of anaerobic bacteria)). If your tank water has the smell of rotten eggs, that is a sign of anaerobic waste.

The best thing you can do for your fish right now is to vac your gravel, fill your tank, and then vac it again untill you clean it all.

Also, have you tested your water? What are the parameters?

i want to thank you guys 4 taking out the time 2 help me:headbang2

On another note, it's good that you have carbon in your filters. Excellent for removing impurities and for polishing the water, however if you've had it in your filters for more than a month, it's probably not doing its job very well. Replace with new carbon if your existing carbon is old.
