If your keeping hardy fish then adding de-chlor while your python is filling is fine. If your keeping rummynose tetras or cardinals who don't handle fluctuation well I would prepare your water prior to adding.

See I've been preparing my water ahead of time "aging" which I guess is fine but I feel like it takes forever. Thanks for your help guys. And thanks for the link sick_lid.
