Will you be setting up the new tank same day you take down the old?
Keep your bio media to help get you started when you set your tank back up. There's no benefit to keeping any old tank water, other than to store your bio media in during the move, in a bucket or something.
I will also say that hell of a lot of good BB will be in your substrate. If you are replacing all your substrate with new be very careful, the chances are you will have an imbalance of BB when you set your tank back up.
This could skew your parameters until things settle down.
You'll need some kind of oxygenation so yes an air stone or sponge filter to keep your fish and bb aliveDuring the transportation I am going to put the bio media into a bucket with the tank water but do I need to run a probable pump for them?

Not if it's a short trip.. but if its gonna takes hours to move the fish then probably a good idea ?I mean do I need to have a portable pump running in the bucket during the transportation? When the fish and the bio media arrive at the new place I will put them all together into a storage box with my Eheim 2215 running
