I add the full shot of Prime (1 cap for each 75gal tank with a 50% water change) right when I begin adding the temperature adjusted tap water. Never had any problems doing it this way over the years.

I do 50% water changes.

I dose an amount equal to the whole tank, before I start re-filling. That way, the current from filling will mix it around with the new water.

Never had any issues.

You really want your dechlorinator in the tank before you add the fresh tap water, that way it is available to reduce the chlorine as it enters the tank. But a few minutes with diluted chlorine containing water probably won't really harm the fish too much.

I usually do 1/2 the dose as i'm refilling and the other 1/2 about halfway or so during the refill process. sometimes i'll do the full dose at the begining.. but never after the tanks been refilled.

1/2 the dose as I start refilling the tank, 1/2 again just before I finish filling and water starts falling into the sump.
 
I dose the full tank volume then fill with water been doing it for a year never had any issues.
