No the new silicone won't stick to old. I've found that if it's new recently dried silicone it will stick. if you need to patch a spot or just want to add extra it will stick.

I have used GE1 to build many tanks over the years. I use it on smaller to medium size aquariums with no problems. I have a few tanks that are approaching 15 years old.
For a large tank anything over say 125 or 150 I recommend this: http://www.grainger.com/product/MOMENTIVE-Silicone-4UH12
I used this silicone on a 220 about three years ago and it is holding strong.
As far as sealing over old silicone. New will not adhere to the old, If it is a small leak and the rest of the silicone in the tank is in good shape you could try to use a bead of silicone just to fill the spot and stop the leak.
You will get no strength from this. The silicone between the glass is what holds the tank together. The bead inside adds very little strength and mainly just provides the water tight seal.
The best way is to completely take the tank apart and clean all the old silicone off and reassemble.

GE 1 window and door is not toxic to fish. I never used GE 2

GE 1 is safe. GE 2 is toxic. Anything that is anti mold/mildew will leach chemicals. Stick to GE 1, which has blue writing

Thanks for all of the advise guys. I'm going to tackle this today. Luckily I have a spare tank in the garage that will hold my fish for a week or so while I do this. I've read a lot into soil substrates and I believe I'm going to pass this time around. From what I've read it's best to only keep the water only two or three inches above the soil and sand for about two months before you fill that tank. I just can't afford that with my fish needing to go back in ASAP. I will keep every one posted through the weekend. Thanks a gain everyone.
